曲克芦丁减轻BDE-47诱导的小鼠肝脏氧化应激

摘要目的  利用四溴联苯醚(2,2′,4,4′-tetrabromodiphenyl ether,BDE-47) 建立小鼠肝脏损伤模型,研究曲克芦丁对BDE-47诱导的小鼠肝脏氧化应激的保护作用。方法  建立BDE-47诱导的小鼠肝脏损伤模型,用曲克芦丁处理,设置正常Control组、BDE-47 组、BDE-47(150mg/kg/day)+Troxerutin (150mg/kg/day)组和Troxerutin组共四组,连续喂养12周后处死。以生化检测的方法检测小鼠肝脏中ALT的含量、ROS含量、4-HNE含量、GPX和SOD酶的活力和GSH含量。结果BDE-47能够引起小鼠肝脏细胞中ALT含量升高、ROS与4-HNE含量增加、GPX和SOD酶的活力降低和小分子抗氧化物GSH含量下降;曲克芦丁能使ALT、ROS、4-HNE、GPX、SOD和GSH恢复到对照组水平。而且单独加入曲克芦丁对于ROS与4-HNE含量,GSH含量,GPX和SOD酶是没有影响。结论  曲克芦丁能够有效地减少由于BDE-47所引起的ROS与4-HNE含量提高与GSH含量的减少,并且对于BDE-47所引起的GPX和SOD酶的活力降低的现象能够有效地改善。69174

Troxerutin attenuates oxidative stress induced by BDE-47 in mouse liver 

Abstract Objective: In this study, we established the mouse model of liver injury to research the protective effects of troxerutin on BDE-47-induced hepatic oxidative stress. Method: 24 mice were pided into 4 groups at random:control group, BDE-47 group, BDE-47 150mg/kg/day + Troxerutin 150mg/kg/day group and Troxerutin group. After 12 weeks of treatment, mice were sacrificed to determine the serum level of ALT, ROS and 4-HNEcontent, GPX and SOD enzyme activities, as well as GSH content in the livers of mice. Results: BDE-47 markedly elevated serum ALT level and hepatic ROS and 4-HNE content, decreased the hepatic activities of SOD and GPX and GSH content in mice. However, troxerutin significantly reduced serum ALT level and hepatic ROS and 4-HNE content, increased the hepatic activities of SOD and GPX and GSH content in BDE-47 treated mice. Conclusion:  Troxerutin could effectively protect mouse fromBDE-47-induced liver injury by attenuating ROS and 4-HNE content, increasing the activities of SOD and GPX and GSH content.
