曲克芦丁抑制BDE-47诱导的小鼠肝脏细胞凋亡

探讨曲克芦丁对BDE-47诱导的小鼠肝脏细胞凋亡的保护作用和分子机制。方法:建立BDE-47诱导的小鼠肝脏损伤模型,再用曲克芦丁(troxerutin)处理,设置Control组、BDE-47(150mg/kg/day)组

摘要目的:利用四溴联苯醚(2,2′,4,4′-tetrabromodiphenyl ether,BDE-47) 建立小鼠肝脏损伤模型,本文探讨曲克芦丁对BDE-47诱导的小鼠肝脏细胞凋亡的保护作用和分子机制。方法:建立BDE-47诱导的小鼠肝脏损伤模型,再用曲克芦丁(troxerutin)处理,设置Control组、BDE-47(150mg/kg/day)组、BDE-47(150mg/kg/day)+ Troxerutin (150mg/kg/day)组和Troxerutin(150mg/kg/day)组共四组,连续喂养12周后处死。(1)以生化检测技术对小鼠肝脏氧化应激指标进行检测。(2)以Western blot法对小鼠肝细胞凋亡相关通路指标进行检测。结果:(1)BDE-47处理后小鼠肝脏细胞中自由基含量明显升高,曲克芦丁处理后使得肝脏中自由基含量明显减少。(2)BDE-47处理后小鼠肝脏细胞内质网中P-PERK、P-eIF2含量显著升高,曲克芦丁处理后使得肝脏细胞内质网中P-PERK、P-eIF2含量明显减少。(3)BDE-47处理后小鼠肝脏细胞凋亡相关蛋白Cleaved-Caspase-3、GADD34和Bax含量均显著升高,抗凋亡蛋白Bcl-2含量明显减少。曲克芦丁处理使得小鼠肝脏细胞凋亡相关蛋白Cleaved-Caspase-3、GADD34和Bax含量明显减少,抗凋亡蛋白Bcl-2含量明显升高。结论:曲克芦丁能够通过抑制BDE-47诱导的小鼠肝脏氧化应激,进而抑制肝脏内质网应激从而抑制肝脏细胞凋亡。50725

Troxerutin Inhibits BDE-47-induced Apoptosis in Mouse Liver

 Abstract 

Objective:In this study, mouse model of liver injury induced by 2, 2 ', 4, 4 '- tetrabromodiphenyl (BDE -47) was established to explore the protective effects of troxedrtutin on apoptosis in mouse liver. Methods: 24 mice were pided into four groups: Control group, BDE-47 group (150mg/kg/day), BDE-47(150mg/kg/day) +Troxerutin group (150mg/kg/day)and the Troxerutin (150mg/kg/day). After 12 weeks of treatment, mice were sacrificed, then serum ALT level, hepatic ROS level and the protein levels of P-PERK, P-eIF2, Cleaved Caspase-3, the Bcl-2 and GADD34 were determined.Results: BDE-47 treatment induced a significant increase in the levels of serum ALT, hepatic ROS and the protein levels of P-PERK, P-eIF2, Cleaved Caspase - 3 and GADD34, and a notable decrease in Bcl-2 level in mice. Nevertheless, troxerutin markedly decreased the levels of serum ALT, hepatic ROS, the protein levels of endoplasmic reticulum stress markers and apoptosis-related protein, and restored the protein level of Bcl-2 in BDE-47-treated mice. Conclusion: Troxerutin could protect against BDE-47-indued apoptosis via inhibiting oxidative stress, thereby attenuating ER stress, furthermore suppressing apoptosis-related signaling in mouse liver.
