摘要由于太空飞行环境的特殊性,研究发现其对航天员的免疫系统会产生一些不利的影响,比如说太空环境可以造成太空飞行者被细菌、真菌和病毒感染的几率增加,并增加航天员患自身免疫性疾病的风险,并且空间飞行能导致生物体免疫系统多种效应的改变及潜伏病毒的活化。本文以从人外周血中获得纯化的T淋巴细胞为研究对象,探究其在RCCS旋转细胞培养系统产生的模拟微重力条件下,T淋巴细胞细胞增殖活力和细胞因子IL-2表达的变化。本实验中细胞增殖活力的检测使用了CCK-8法,IL-2的检测主要使用了酶联免疫吸附法。实验得到的结果表明,模拟微重力条件抑制了T淋巴细胞的增殖活力;研究证实在刀豆蛋白的刺激下,T淋巴细胞IL-2分泌水平升高,而模拟微重力条件抑制了T淋巴细胞IL-2的分泌,也抑制了刀豆蛋白刺激条件下T淋巴细胞IL-2的分泌。30467

Title  The change of the function of T lymphocyte in human peripheral blood on simulated microgravity condition                                      
Abstract
Some reports show that space flight can have negative effects on the astronauts’ immune system, it leads to bacterial, fungal or viral infections, and increase the risk of autoimmune disease. Space flight also lead to  organisms’ immune system change and the latent virus to be activated.The purified T lymphocyte from human peripheral blood is the research object of this article . We studied the comparison of cell vitality and the expression change of  IL-2  under different culture conditions including the rotary cell culture system (RCCS) under the simulated microgravity conditions and the horizontal conditions.The method we used to detect the T cell’s vitality was the CCK-8 . And the method to test the IL-2 content was the enzyme-linked immunosorbent assay. The results show that under the microgravity condition, the vitality of T cell and IL-2 expression changed, microgravity conditions significantly reduce the vitality of T cell and inhibit the production of IL - 2 when the ConA’s stimulation exist or not.
