水蒸气蒸馏法八角风味物质的高效提取工艺优化

摘要:八角茴香,是木兰科八角属植物,主要分布于我国广西、广东、云南等省区,是我国南方一种重要的“药食同源”的作物。作为一种常见的烹饪佐料,直接取用八角茴香果实进行烹饪,无法完全利用八角茴香中的各种成分,因而对八角茴香的各成分进行提取、精炼、分类,将能够极大地提高八角茴香的利用率。  将八角茴香果实进行粉碎,晾干。采用两种方法对八角茴香风物质进行提取,首先采用索氏提取法提取八角茴香中的油脂类物质,以单因素实验对提取条件进行选择,依次比较提取溶剂,提取温度,提取时间,样品粒径大小对提取效率的影响。最终确定索氏提取法的最优工艺条件为:丙酮作提取剂,95℃提取,80目粒径,4小时提取时间为最佳。
再用水蒸气蒸馏法,采用挥发油测定器,进行样品的挥发油提取。以单因素实验对提取条件进行选择,依次比较提取时间,料液比,样品粒径大小,预浸泡时间对提取效率的影响。最终确定水蒸气蒸馏法的最优工艺条件为:提取 2小时,料液比1:30,20目粒径,预浸泡 72小时,且进行冷冻预处理,样品挥发油得率最高。22578
实验最后采用 GC-MS 对提取物进行分析,比较两种提取方法下的八角茴香挥发油主要成分的含量。

Efficient extraction process optimization of
Illicium Verum Hook .f flavor  
Abstract:  Illicium Verum Hook .f  is a species of Illicium in Magnoliaceae  which
mainly grows in Guangxi, Guangdong, Yunnan and some other provinces in China.
It’s an important kind of medicine food homology crop in southern China. As a kind
of common seasoning,  the ingredients in illicium Verum Hook .f can’t be fully used
when it’s cooked directly. But through the process of extracting, refining and
classifying,these ingredients can make their best uses.
Shatter and dry the illicium Verum Hook .f sample and use two ways to extract its
ingredients. First, use the Soxhlet extraction method to extract grease-like material.
Do single factor experiment to choose extraction conditions. The factors include the
solvent, the temperature, the extracting time and the partical size of sample in the
process of extraction. From the result we can see the best condition is using the
acetone as solvent, controlling the temperature in 95℃. The partical size of sample
should be at 80 mesh and 4 hour is enough for the extraction.
Then, use the steam distillation method with volatile oil determination apparatus
to extract the essential oil. Do single factor experiment to choose extraction
conditions. The factors include the extracting time, the material-liquid ratio, the
partical size of sample and the time that the sample is soaked. And the best condition
is: 2 hours’extraction, a 1:30 material-liquid ratio, partical size of 20 mesh and 72
hours’ soaking time. If the sample is frozen and then thawed, the result of extraction
will be better.
At the end of the experiment, we use  the GC-MS to analyze the  extractive,
comparing the main ingredients of the essential oil in illicium Verum Hook .f extracted
through the two methods.
