斑马鱼LRRK2突变体中关于小儿多动症和多巴胺系统相关基因的qRT-PCR分析

摘要大多数学者认为注意力缺陷多动障碍的发病因素是由多方面因素造成的神经生化以及精神药理学相关的研究发现,多动症患者的血和尿中多巴胺和去甲肾上腺素功能低下。故而,有学者提出注意力多动症ADHD可能与多巴胺的多少存在某种关系。我将就其中的一种假说神经递质来进行探究。目前为止,在生物研究领域,大多数学者业已认为LRRK2基因是帕金森综合症的治病基因,帕金森综合征患者体内检测到多巴胺的减少。我们也发现在小儿多动症的模型中多巴胺也是有所减少的。所以,有学者提出多巴胺的减少有可能与小儿多动症的发病有关。于是,研究者便做了一系列相关的实验,以斑马鱼为模式动物,以斑马鱼LRRK2基因的突变体和斑马鱼的野生型为相关的对照组,探究出LRRK2基因的确能够在一定程度上影响体内多巴胺细胞的产生,从而,在一定程度上,和小儿多动症的发病存在某些相关性。我们也因此取养殖斑马鱼的LRRK2基因突变体与斑马鱼野生型的6天 的卵提取RNA;多巴胺相关的的基因lphn3.1以及betaactin基因以qrt-pcr技术来验证是否在斑马鱼LRRK2突变体中存在某种联系。实验结果表明:lphn3.1基因在野生型斑马鱼的表达明显高于lphn3.1基因在Lrrk2突变体斑马鱼的表达。第一次在帕金森的模型里找到与多动症模型表达相关的基因。49353

However, recently, some scholars put forward LRR2 gene may be associated with the pathogenesis of adhd。Therefore, the researchers made a series of relevant experiments, zebrafish as model animals, in zebrafish LRRK2 gene mutant and wild type of zebrafish for relevant control group, explore the LRRK2 gene does to a certain extent, affects the production of dopamine in the body, thereby, to a certain extent, and the onset of adhd from in some correlation. Thus we collect breed zebrafish LRRK2 gene mutant and wild type of zebrafish 6 days the eggs of extracting RNA; Dopamine lphn3.1 related genes and genes betaactin QRT - PCR technology to verify whether there is a link between in the zebrafish LRRK2 mutants.The experimental results show that: Lphn3.1 gene significantly better than in the expression of wild-type zebrafish lphn3.1 genes expressed in Lrrk2 mutant zebrafish. This is consistent with the researchers' experimental results, for the first time found in Parkinson's disease model with adhd model expression of related genes.
