高粱SbFtsH基因的分子克隆表达及进化研究

摘要:FtsH基因家族是一种广泛存在于原核和真核生物中的ATP依赖型金属蛋白酶。本文以拟南芥的FtsH基因为检索序列,利用BLASTP程序对高粱的基因组进行扫描,得到在高粱基因组中的10个FtsH基因。序列表达分析,Sobic.003G195000和Sobic.009G256500序列是一对片段重复序列,Sobic.009G256500序列表现为特异型表达,而Sobic.003G195000序列则表现为组成型表达。基因的结构与保守基序特征分析,FtsH基因家族的结构可分为七类,保守蛋白包括三种保守基序。系统进化分析,10个FtsH蛋白成员可分为6个类群,其中Sobic.003G195000和Sobic.009G256500序列是一对特异同源序列。组织表达谱分析,在46个高粱器官组织或处理条件下Sobic.003G195000序列的FPKM范围为2.43~4.91,Sobic.009G256500序列的FPKM范围为-10~-5.03,Sobic.009G256500序列在表达上发生了分化,表现为表达上的多样化。从基因结构和保守蛋白两方面分析,Sobic.003G195000序列和Sobic.009G256500序列的基因发生的突变低。本研究为进一步探明高粱FtsH 基因家族成员的生物学功能提供理论指导。43753

Molecular Cloning, Expression and Evolution of SbFtsH Genes in Sorghum

Abstract: FtsH gene family is a kind of ATP dependent metalloproteinase, which is widely existed in the prokaryotic and eukaryotic organisms. In this paper, the sorghum genome was scanned using Arabidopsis FtsH gene sequences as queries, and using the blastp program. A total of 10 FtsH genes were identified in the genome of sorghum. Sequence and expression analysis showed that Sobic.003G195000 and Sobic.009G256500 is segmentally duplicated, and that Sobic.009G256500 and Sobic.003G195000 was constitutively expressed. Analysis of gene structure and conserved motifs, the structure of the FtsH gene family can be pided into seven categories, including three conserved protein motifs. Phylogenetic analysis, 10 FtsH protein members can be pided into 6 groups, including Sobic.003G195000 and Sobic.009G256500 sequence is a pair of specific homologous sequences. Tissue expression profile analysis, the Sobic.003G195000 sequences of 46 sorghum organs or processing conditions in the FPKM range of 2.43~4.91, the Sobic.009G256500 sequence of the FPKM range of -10~-5.03, Sobic.009G256500 had the differentiation in sequence the expression, for the performance of the persification on expression. Analysis from two aspects of gene structure and conserved protein, Sobic.003G19 The gene mutation of the 5000 sequences and Sobic.009G256500 sequences was low. This study provides theoretical guidance for the further study of the biological function of the members of the FtsH gene family in sorghum.
