聚谷氨酸/壳聚糖水凝胶的制备及性能表征

摘要:本实验分别从接种量、含水量、氯化钠浓度、镁离子浓度、磷酸氢二钾浓度等一系列因素进行单因素试验,找出各因素最优值对γ-PGA的固体发酵培养基进行优化。其结果表明接种量、氯化钠浓度、磷酸氢二钾浓度为γ-PGA产量的主要影响因素。接种量为1.4ml,氯化钠的浓度为0.78%,磷酸氢二钾的浓度为0.003mmol/g。在此条件下γ-PGA的产量为0.1915g,比优化前γ-聚谷氨酸产量提高了6.5%。然后进行了壳聚糖/水凝胶的制备及性能表征,结果表明在37℃时聚谷氨酸/壳聚糖水凝胶有较好的溶胀性能,并且在中性条件下的溶胀率最好,饱和溶胀率达到120%.溶胀平衡的水凝胶在酸性条件下消溶胀的速度比中性条件下的溶胀速度更快。并且就较好的生物降解性,再37℃下用1mg/ml的木瓜蛋白酶40小时的降解率达到58%。23475

TPGA / chitosan hydrogel preparation and characterization
Abstract: In this experiment, respectively from the inoculation amount, water content, the concentration of sodium chloride, magnesium ion concentration, the concentration of potassium hydrogen phosphate two and a series of factors of single factor test, analyzed the factors of optimal value of medium optimization of solid fermentation of γ -PGA. The results showed that the inoculation amount, concentration of sodium chloride, potassium hydrogen phosphate two concentration was the main factor influencing the production of γ -PGA. The inoculation was 1.4ml, the concentration of sodium chloride was 0.78%, concentration of potassium hydrogen phosphate 0.003mmol/g two. Under these conditions, the yield of 0.1915g γ -PGA, than before optimization of gamma polyglutamic acid production increased by 6.5%. Then the chitosan / hydrogel preparation and characterization, results indicated that in 37 ℃ PGA / chitosan hydrogel has good performance and swelling, swelling in  
the neutral condition was the best, the saturated swelling rate of 120%. equilibrium swelling of hydrogel in acidic conditions deswelling faster than the swelling rate of neutral under the conditions of. And good biodegradability, degradation and 37 ℃ with 1mg/ml papain 40 hour rate reached 58%.
