摘要：卡非佐米是一种新一代的蛋白酶抑制剂。主要用于治疗多发性骨髓癌。本次实验主要研究的是卡非佐米关键中间体三肽的制备方法以及工艺优化，着重研究了每步反应的要点并进行了改良，以达到较优的制备方案。最终确定了以L-苯丙氨酸甲酯盐酸盐，Boc-L-亮氨酸与Boc-L-高苯丙氨酸为原料，分别经过脱水缩合、亲核取代、酰氯化，水解等一系列反应来制备目标产物的合成路线，同时在实验过程中对投料比、温度等进行了研究，以此确定了较好的工艺条件。通过一系列的工艺优化最终确定了一条产率和纯度较高、操作较简便的工艺路线。22471

Study on the synthesis of the key intermediate three peptide of Carfilzomib
Abstract：Carfilzomib is a new generation of protease inhibitor. It is mainly used for the treatment of multiple myeloma. The experiment is mainly study the preparation method and process optimization of the key intermediate three peptide to Carfilzomib. This paper focuses on the key points of each reaction and improved them, in order to achieve the optimal preparation scheme. The final determination of the synthetic route which use Methyl L-phenylalaninate hydrochloride, N-Boc-L-leucine and boc-L-homophenylalanine as raw material, after dehydration condensation, nucleophilic substitution, chloroformylation reaction, hydrolysis reaction and a series of reaction to prepare the target product. At the same time, in the course of the experiment, the feed ratio, temperature and so on, were studied in order to determine the better techniques. By the process optimization, a high yield, high purity and simple operation of process route was determined.

1.1卡非佐米简介
